Optogenetics-rAAV

Optogenetics is a technology that is largely used to manipulate neuronal activity. AAVs are used to drive the expression of light-activated ion channels in select neurons. Fiber optics are then directed toward the cells that express these light-activated ion channels, and light can be applied to rapidly manipulate the influx of charged ions and subsequently drive changes in neuronal activity. Most commonly, the ion channel Channelrhodopsin-2 (ChR2) is used to activate neurons while the ion channel Halorhodopsin (NpHR) is used to inhibit neurons. This technology provides a mechanism for selective manipulation of neurons at precise timescales during physiological or behavioral experiments and serves as a powerful tool for neuronal circuit mapping.
