Optogenetics is a technology that is largely used to manipulate neuronal activity. AAVs are used to drive the expression of light activated ion channels in select neurons. Fiber optics are then directed toward the cells that express these light activated ion channels, and light can be applied to rapidly manipulate the influx of charged ions and subsequently drives changes in neuronal activity. Most commonly, the ion channel Channelrhodopsin-2 (ChR2) is used to activate neurons while the ion channel Halorhodopsin (NpHR) is used to inhibit neurons. This technology provides a mechanism for selective manipulation of neurons at precise timescales during physiological or behavioral experiments and serves as a powerful tool for neuronal circuit mapping.
