Cap 1 N1meψ SpCas9HF mRNA

SpCas9HF, also known as High-Fidelity SpCas9, is a variant of the CRISPR-associated protein Cas9 derived from the bacterium Streptococcus pyogenes. This variant has been engineered to reduce off-target effects, enhancing its specificity for target DNA sequences. The “HF” designation indicates its high-fidelity nature, meaning it exhibits improved accuracy in discriminating between target and non-target DNA sequences. SpCas9HF retains the ability to target specific DNA sequences and induce double-strand breaks, like the wild-type SpCas9, but with reduced off-target effects, making it valuable for applications where precision is critical, such as gene therapy and genome editing. This mRNA possesses a Cap1 structure, which is highly efficient in capping. It is fully substituted with N1-methyl-pseudo Uridine to improve expression and decrease immunogenicity. Additionally, the mRNA features a 110A tail in its sequence, resembling a mature mRNA.
