Greenland Mines Receives Canadian Patent for Klotho-Based Gene Therapy Technology

Greenland Mines announced that Innovation, Science and Economic Development Canada has granted Canadian Patent No. CA3,005,398, titled “Secreted Splicing Variant of Mammal Klotho as a Medicament for Cognition and Memory.”

The patent covers technology exclusively licensed for worldwide commercialization from researchers at Universitat Autònoma de Barcelona and collaborating institutions. It protects the use of a human gene therapy based on a nucleic acid sequence encoding the RNA splice variant protein known as secreted Klotho, or s-KL.

According to the company, the issued patent includes 42 granted claims covering s-KL, methods of gene delivery, protein synthesis from the gene, and the potential use of s-KL-based genetic therapy in several neurodegenerative and neurological conditions.

Klotho is a protein associated with aging biology and has been studied for its roles in cognition, metabolism, kidney function, and neuroprotection. Greenland Mines said its Klotho-based therapeutic programs are designed to restore Klotho protein levels toward normal tissue concentrations in the brain, heart, kidneys, and other organs.

The company stated that age-associated epigenetic changes may reduce Klotho expression over time, contributing to biological processes linked to aging. Its therapeutic strategy is intended to increase tissue Klotho levels as a potential approach to preventing or treating diseases associated with aging-related dysfunction.

The Canadian patent specifically supports KLTO-101, Greenland Mines’ product candidate involving secreted Klotho. The company said the patent provides protection across multiple potential indications, including Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, ALS, Lewy body dementia, multiple sclerosis, ataxia telangiectasia, post-stroke dementia, traumatic brain injury-related dementia, senile dementia, and craniocerebral trauma.

Although the announcement does not specify the exact delivery vector for KLTO-101, Klotho gene therapy programs may be relevant to the broader AAV and genetic medicine field, where durable in vivo delivery is commonly explored for neurological and systemic disorders. Key development questions will include vector selection, tissue targeting, dose, durability of expression, and safety.

Greenland Mines said the Canadian patent expands its global intellectual property position for KLTO-101, adding to issued patents in major markets including the United States, China, and Europe.

The company plans to begin process development and manufacturing for KLTO-101 in August 2026. Greenland Mines also stated that KLTO-202, a separate Klotho-based program for ALS, multiple sclerosis, and other neuromuscular disorders, has advanced to the GMP manufacturing stage.

The company expects to begin interactions with the FDA and other regulatory authorities before the end of 2026. As the programs move forward, key questions will include delivery strategy, potential AAV applicability, tissue-specific expression, dose selection, safety profile, and clinical validation of Klotho modulation in neurodegenerative and neuromuscular diseases.
