Early Phase 1 Results for BAFF-R-Targeted CAR T Show Durable Complete Responses in R/R B-Cell Lymphoma, Including After Prior CD19 CAR-T Failure

PeproMene Bio has announced the publication in The Lancet of Phase 1 results for PMB-CT01, a first-in-class BAFF-R-targeted chimeric antigen receptor (CAR) T-cell therapy, in patients with relapsed or refractory B-cell non-Hodgkin lymphoma.

Seven of nine treated patients, or approximately 78%, achieved a complete response, including four of six who had progressed after conventional CD19-directed CAR T-cell therapy (“CAR after CAR”). All complete responses remained ongoing at data cutoff, with no relapses observed and the longest ongoing response reaching 35 months.

The therapy was reported to be well tolerated: all cytokine release syndrome (CRS) events and the two reported immune effector cell-associated neurotoxicity syndrome (ICANS) events were limited to Grade 1. Based on these results, the Phase 1 trial has expanded from City of Hope to additional U.S. medical centers.

By targeting BAFF-R rather than CD19, the approach offers a potential option for patients who relapse after existing CD19-directed CAR T-cell therapies.
