AAV titer quantification by ddPCR typically starts with sample preparation: capsid lysis followed by DNase I digestion and heat treatment effectively releases the viral genomes. Next, during droplet generation, the PCR reaction mixture is precisely partitioned into approximately 20,000 nanoliter-sized, water-in-oil droplets using automated droplet generator. In the amplification phase, target sequences (such as AAV ITR regions) are PCR-amplified, with fluorescent probes specifically marking positive droplets containing AAV genomes. Finally, quantification is achieved by analyzing droplet data with Poisson statistics to determine the absolute concentration of genome copies per microliter.
